The Haynes manual is correct, not sure which model of the bike you have but you need to check out the correct manual from here

I looked at the K3 pointy manual on page 8-38 (Inspection of the speed sensor) and it describes the test method. Its a hall effect magnetic sensor and cant be measured just by connecting a multi-meter to the wires.
The batteries are to provide voltage to the circuit when you have the speed sensor disconnected from the wiring loom, you may find it easier with a 9V battery as its easier to connect and wont go rolling around the floor.
When its rigged up you should spin the wheel slowly and see the voltage measured vary from 0v and whatever voltage battery you use e.g. 9v. If thats working then the sensor may well be fine. Check for obvious damage though by removing the front wheel, the little lugs tend to get chewed off preventing the rotor from rotating properly.
If you can get one, far easier to just plug in another speedo and spin the wheel. Make sure that you actually have power to the speedo unit (are the warning lights coming on etc) otherwise nothing will work. Other things to check are obvious poor connections, particularly where the speedo connects to the loom, give that a squirt of WD40. And of course take a look at the fuses and make sure they are all intact, especially the one for the signal circuit as this powers the speedo unit on the later models.
